中文摘要: 速激肽对哺乳动物繁殖有重要调节作用,山羊的繁殖具有明显的季节性,是建立长日照信号理想的实验动物模型。本研究采用RACE技术克隆速激肽及其受体基因全长cDNA序列;利用qRT-PCR、Western印迹和免疫组化技术分析速激肽及其受体基因在山羊下丘脑垂体、卵巢等组织中的表达情况;测定光照信号刺激对垂体、卵巢等组织中速激肽及其受体基因表达变化和血液中生殖激素的影响和在体外培养条件下速激肽对垂体细胞促乳素分泌能力的影响,研究速激肽基因作用垂体细胞调节促乳素释放的信号通路。通过研究,旨在揭示速激肽及其受体基因作为长日照信号诱导生殖激素释放、调节山羊生殖规律的机理,同时对深入理解人为时钟信号和组织内在时钟信号调控季节性繁殖的哺乳动物垂体激素释放机理提供新的思路,为速激肽及其受体基因在山羊遗传育种和繁殖领域的深入研究提供理论依据。

英文摘要: Tachykinins play an important role in the control of mammal reproductive functions.Seasonally breeding animals such as goat is an ideal animal to establish long-day experimental animal model.In this study, the Race technique would be used to clone the Tachykinins(TKs) and tachykinin receptors(TKR) in goat.Then,the Quantitative real-time by PCR,western blot and immunohistochemistry would be used to test the differential expression of Tachykinins and tachykinin receptors on the hypothalamus, pituitary and ovarian tissue,and to study effects of tachykinins on prolactin secretion in vitro,and to detect the signaling pathway of regulating prolactin-releasing by TKs.This reseach aims to reveal the mechanisms of tachykins as long-day signals inducing seasonal hormone changes and regulating reproduction in goats.The research of TKs and TKRs in genetic breeding of goat offers important new insight into the complex mechanisms controlling pituitary hormone release by timed signals and the internal biological clockwork in this tissue,and provides theoretical basis for further studying of TKs and TKRs in goats genetic breeding and reproduction.
