中文摘要: 胚胎植入是生殖过程的一个关键步骤,与肿瘤发生发展有许多相似生理特性,因此对其分子机制的了解将有助于生育调节技术的发展和肿瘤防治技术的探索。NDRG家族基因广泛表达于对细胞应激信号产生反应的组织器官,并在肿瘤的发生发展中发挥重要的作用。NDRG1和NDRG3是该家族的2个成员,虽然目前对于它们在胚胎植入过程中的功能及其作用机制尚无报道,但我们在前期研究中观察到,NDRG3的表达受雄激素的调控,并与肿瘤细胞的迁移特性相关,而且NDRG1和NDRG3在小鼠围植入子宫内膜组织的表达都呈特异性变化,提示它们可能参与调控小鼠胚胎植入过程。本项目拟在前期工作基础上,从其在小鼠子宫内膜组织中的表达模式和上下游调控途径入手,研究分析NDRG1和NDRG3在胚胎植入中的作用机制,筛选鉴别新的生物标志分子,以期加深对胚胎植入分子机制的了解,并为揭示不孕不育的发生机制及其临床诊治技术的发展提供科学依据和物质基础。

英文摘要: As a key event during reproduction, blastocyst implantation is a highly regulated process. There are striking similarities present between the behavior of invasive embryonic trophoblast cells and that of invasive cancer cells. A better understanding of the mechanism of embryo implantation will likely lead to better insights of various diseases of pregnancy, and lead to better understanding of metastatic cancer cells. The N-myc downstream-regulated gene (NDRG) family consists of four proteins: NDRG1, NDRG2, NDRG3, and NDRG4 in mammals, and has been thoroughly studied as an intracellular protein associated with stress response, cell growth, and differentiation. It has been reported that NDRG1 involved in the pathology of cancer. Till now, detailed mechanisms of NDRG in blastocyst implantation are still poorly understood. Our preliminary study demonstrated that NDRG3 is significantly upregulated by androgen and contribute to the malignant phenotype of tumors. Out results in mice uterus also showed that the temporal and spatial expression of NDRG1 and NDRG3 was highly coincidence with the mice embryo implantation, which suggested that NDRG1 and NDRG3 play an important role in implantation.
